After completing his high school education,. Medupe decided to pursue a National Diploma in Drama from the Tshwane University of Technology. So, he graduated in 2010, after 3 years of studying for the degree in 2007.

Unlike his gay role on Smoke and Mirrors, JT Medupe is not gay in real life, but a husband and father. As of 2024, JT Medupe is dating fellow actress Lebogang Mthunzi, whom he shared a screen with on Generations as her brother. JT Medupe played the role of Jonathan Motene on the show, while Lebogang Mthunzi played Angela Mogale.

When Medupe was one year away from completing his tertiary education, he joined the Contractors Artists management agency. And through them, he secured a lot of TV roles. However, acting is not all that Medupe did over the years.
From 2013 to 2014, he actually produced and wrote for MTTV, which produced telenovelas such as Scars. Thereafter, between 2014 and 2015, he was a casting assistant and a production assistant for Ferguson Films.
Furthermore, he has also freelanced for Blazaai Pictures from January 2018 to November 2019. In terms of acting, he’s more famous for his role on Generations: The Legacy as Jonathan Motene.
